An electron microscopic study on rat liver after INH administration

Abstract
The effects of isoniazid on fine structure of hepatic cell were studied in the rats. Rats were received, daily, 0.8mg(usual dose) and 3.2mg(large dose) per 1008 body weight of INH, respectively, through gastric intubation, and were sacrified 2, 4, 6 and 8 weeks, after administration of the drug.
1. Epon toluidine blue stained sections showed well preserved hepatic cell cords and occasional a few fat droplets in the hepatic cells, which were not changed significantly according to either the dose of the drug or the duration of the drug administration. From 4 weeks to 8 weeks after the drug administration a few degenerating cells were present and some inflammatory cells such as neutrophils, eosinophils, lymphocytes, and plasma cells were noted mainly in the interlobular connective tissues.
2. Electron microscopy on hepatic tissues from INH treated rats confirmed the light microscopic findings and revealed the detailed fine structures of hepatic cells. Glycogen contents generally, appeared to be normal except for slight decrease in some instances notably after 6 weeks and 8 weeks. Smooth endoplasmic reticulum was often noted in close association with glycogen area and its amount appeared to have a reverse relationships with that of glycogen particles. Granular endoplasmic reticulum were generally well preserved, but same were irregularly arranged and showed often cisternal dilatation and partial detachment of ribosomes. Fat droplets were occasionally found in hepatic cells especially in their basal regions. Bile canaliculi were noted to be somewhat dliated and sometimes contained dense materials or cell debris in the lumina, more often from 6 weeks. From six weeks, collagenous fibers appeared to increase in the Disse spaces and were often present as thick bundles. A few degenerating cells having dense bodies, vesicular endoplasmic reticulum, and swollen mitochondria were present especially from four weeks. There were inflammatory cells in the perisinusoidal spaces and more often in the untErlobular connective tissue in later time period of the experiments.
From these results it may be concluded that INH administration dose not cause widespread hepatitis in the rats, however, fine stractural alterations of some components in the liver observed were thought to be due to the toxic effect of INH on the liver.
